- The self-assembly of molecules into supramolecules is ubiquitous in nature at several scales, from single proteins to membrane formation. The self-organization of these structures is driven by non-covalent interactions which follow the direction indicated by the thermodynamic equilibrium. Peptides, made up of short chains of amino acids, are among the most widespread molecules used as starting bricks for obtaining supramolecules. The reason is that they can be obtained by synthesis and engineered at the molecular level. Indeed, the secondary structures of the peptide building blocks can induce different spatial configurations such as α-helices and β-sheets. External parameters such as temperature, PH, peptide concentration, solvent used as well as ion concentrations drive the kinetics of the process, also allowing to block the assembly in metastable configurations distinct from the thermodynamically preferred one.**_The idea of the PhD project is to add _**
**_plasma_**
**_ as a further external control parameter of the assembly kinetics, in order to obtain new choices on the final configuration of the supramolecules._**
- Atmospheric pressure plasma jets interacting with liquids offer a unique environment inducing electric fields, reactive species formation, localized ion supply, or electrochemical reactions. Depending on the APPJ device different condition can be achieve and used to promote aggregation, orientation, reaction of macromolecules. However, the coupling of the assembling processes usually chemically driven with plasma induced external stimuli is challenging. To allow the realization of this process, a unique device with a double frequency power supply capable of maintaining a low process temperature has been developed in recent years. The PhD activity will consist, on one side, of the optimization of the interaction of the plasma with the liquid surface, using both finite element simulations and plasma electrical and optical diagnostics. In particular, the activity will be aimed at reducing the temperature of the plasma jet, increasing its control over the flow of species and the generated fields and reducing the effects of hydrolysis. On the other side the goal will be the understanding of the action of plasma on the assembling. This will be done through the use of different peptides aimed at highlighting the different contributions in the process. For the characterization of the obtained structures, atomic force microscopy, Raman spectroscopy, electron microscopy and photocorrelation measurements will routinely be used.- The understanding of the new process kinetics will offer novel solutions for tissue growth or for the release of drugs exploitable by biochemists.- In this exciting project your tasks include:
- To characterize a specific APPJ device, focusing in particular on electric fields and currents induced on the facing surfaces depending on the conditions.
- To investigate orientation, aggregation, assembling of macromolecules exposed to atmospheric plasma jet stimuli
- Coordination with internal and external partners
- Publication and presentation of research results in relevant journals or at international conferences

- Università degli studi di Padova - Dipartimento di Fisica e Astronomia- UNIPD has a large infrastructure of characterisation equipment. In particular for the project, it will be accessible: Transmission Electron Microscope (TEM) FEI Tecnai G2 with ultramicrotomes; fluorescence and confocal microscopes; AN2000 (2MeV) and CN (7MeV) accelerators (c/o Laboratori Nazionali di Legnaro - INFN) for Ion Beam Analysis and in particular Rutherford Backscattering Spectrometry (RBS).-
- At the DFA, available with direct use within the project: Secondary Ion Mass Spectrometer-SIMS, Scanning Electron Microscope equipped with energy dispersive spectrometer FESEM ZEISS Sigma HD, UV-Vis Spectrometer, µRaman, ATR-FTIR, Atomic Force Microscope, equipment for photoluminescence measurements, both integrated and time-resolved, using continuous and pulsed (ps and ns) lasers tunable in the visible range and a spectrofluorimeter equipped with a Xe lamp, (Grazing Incidence) X-ray Diffractometer, lithography clean room and a magnetron sputtering systems.
